Molecular Detection and Characterization of Some Virulence (Fnba, Hla and Cna) Genes of Staphylococcus aureus Isolated from Clinical Specimen at Madonna University Teaching Hospital (M.U.T.H), Rivers State

Abstract

Staphylococcus aureus (S. aureus) remains an important cause of both community and hospital acquired infections. Its ability to cause disease is closely linked to several virulence genes including fibronectin-binding protein A (fnbA), alpha-haemolysin (hla) and collagen-adhesin (cna). This study characterized the different virulence genes in S. aureus isolated from different clinical samples at Madonna University Teaching Hospital (M.U.T.H), Elele, Nigeria, and molecularly detect the virulence genes. One hundred and six (106) clinical samples (mainly urine, high vaginal swab, end cervical swab, wound swab, sputum, and skin swab) were collected, cultured for isolation and identification of S. aureus using standard bacteriological methods. The isolated S. aureus were confirmed using a molecular technique (16S rRNA). Genomic DNA extraction was carried out on the confirmed S. aureus using boiling method, and the target virulence genes were detected by polymerase chain reaction (PCR) followed by agarose gel electrophoresis. Twenty-six (26) S. aureus isolates (24.5%) were obtained with higher frequency (76.9%) in urine and (69.2%) in females. Among 26 S. aureus isolated, 18 isolates were confirmed to be S. aureus and were analyzed for virulence genes. Fibronectin-binding protein A was detected in 13 (72.2%), hla in 5 (27.8%), while cna was not detected in any of the isolates. There was no significant relationship between the presence of these genes and age, gender or specimen type (p>0.05). This work provides the first molecular data on the occurance of these virulence genes among clinical S. aureus isolates at MUTH. The high frequency of fnbA and moderate frequency of hla, together with the complete absence of cna, suggest that adhesion and toxin-related mechanism may be important in the infections seen at MUTH, especially those involving urinary tract.
